He recorded seven solo tackles during a 34≢7 loss at the Cincinnati Bengals in Week 2. On September 16, 2012, Skrine earned his first career start after Sheldon Brown was demoted to nickelback. Skrine was elevated to the third cornerback on the depth chart after starting cornerback Joe Haden was suspended for the first four games after violating the league's substance-abuse policy. Head coach Pat Shurmur named Skrine the fourth cornerback on the depth chart to begin the season, behind Joe Haden, Sheldon Brown, and Dimitri Patterson.
